    Population structure and genetic diversity of the threatened pygmy newt Triturus pygmaeus in a network of natural and artificial ponds

    Pond physical characteristics (connectivity, hydroperiod) have shown to be highly relevant in explaining species presence, reproductive success, and survival in breeding-pond amphibians. However, few studies have addressed the influence that these factors may have on the genetic variability of pond populations. We examined genetic variation at 11 microsatellite loci in Iberian endemic, the pygmy newt (Triturus pygmaeus), from 58 breeding ponds in the Doñana National Park (Andalusia), including both temporary ponds and artificially deepened ponds that remain wet during the whole year. Temporary ponds are located in the North part of the region where the surrounding habitat-wet meadows-facilitates the connectivity among populations, whereas the deepest ponds (‘zacallones’) are located in the southern edge embedded in a matrix of unsuitable habitat (thickets and dry underbrush). We investigated genetic diversity and structure within and among ponds. Our results show that both regions (Doñana-North and Doñana-South) are well-differentiated and form two main clusters. We found higher genetic diversity within ponds from the North region, which also exhibited a higher degree of genetic admixture in comparison with populations from the southern edge. Although we found an isolation-by-distance pattern within each cluster, it arose due to the effect of a few isolated ponds located on the edge of each zone, suggesting the existence of substantial gene flow between ponds in the core area. According to our findings, landscape’s permeability to movement (pond connectivity) may constitute a more important factor than hydroperiod length in determining the genetic diversity and viability of pygmy newt populations in this area. Although Doñana populations show a good state, more peripheral and isolated populations present a more worrisome condition as a result of fragmentation and thus, require conservation efforts.     We have developed eleven polymorphic micro- satellite loci for the amphibian Alytes dickhilleni using an enriched-library approach. We detected 98 alleles in 50 individuals genotyped (mean number of alleles per locus was 8.91) in two different populations in South East Spain. Expected heterozygosities ranged from 0.324 to 0.891 in one population (Arroyo Guadahornillos) and 0.424–0.909 in the other population (Cueva Paria). The levels of poly- morphism of the developed markers render them readily applicable for population genetic studies of diversity, structure, and migration.Peer Reviewe

    Expression of the chromatin-associated protein HMGA2 correlates with progression, metastasis and therapy resistance in pancreatic ductal adenocarcinoma (PDAC). Hmga2 has also been identified as a marker of a transient subpopulation of PDAC cells that has increased metastatic ability. Here, we characterize the requirement for Hmga2 during growth, dissemination, and metastasis of PDAC in vivo using conditional inactivation of Hmga2 in well-established autochthonous mouse models of PDAC. Overall survival, primary tumour burden, presence of disseminated tumour cells in the peritoneal cavity or circulating tumour cells in the blood, and presence and number of metastases were not significantly different between mice with Hmga2-wildtype or Hmga2-deficient tumours. Treatment of mice with Hmga2-wildtype and Hmga2-deficient tumours with gemcitabine did not uncover a significant impact of Hmga2-deficiency on gemcitabine sensitivity. Hmga1 and Hmga2 overlap in their expression in both human and murine PDAC, however knockdown of Hmga1 in Hmga2-deficient cancer cells also did not decrease metastatic ability. Thus, Hmga2 remains a prognostic marker which identifies a metastatic cancer cell state in primary PDAC, however Hmga2 has limited if any direct functional impact on PDAC progression and therapy resistance

    Electrical properties of individual tin oxide nanowires contacted to platinum electrodes

    A simple and useful experimental alternative to field-effect transistors for measuring electrical properties free electron concentration nd, electrical mobility , and conductivity in individual nanowires has been developed. A combined model involving thermionic emission and tunneling through interface states is proposed to describe the electrical conduction through the platinum-nanowire contacts, fabricated by focused ion beam techniques. Current-voltage I-V plots of single nanowires measured in both two- and four-probe configurations revealed high contact resistances and rectifying characteristics. The observed electrical behavior was modeled using an equivalent circuit constituted by a resistance placed between two back-to-back Schottky barriers, arising from the metal-semiconductor-metal M-S-M junctions. Temperature-dependent I-V measurements revealed effective Schottky barrier heights up to BE= 0.4 eV

    The Herschel Planetary Nebula Survey (HerPlaNS) - a comprehensive dusty photoionization model of NGC6781

    We perform a comprehensive analysis of the planetary nebula (PN) NGC6781 to investigate the physical conditions of each of its ionized, atomic, and molecular gas and dust components and the object's evolution, based on panchromatic observational data ranging from UV to radio. Empirical nebular elemental abundances, compared with theoretical predictions via nucleosynthesis models of asymptotic giant branch (AGB) stars, indicate that the progenitor is a solar-metallicity, 2.25-3.0 Msun initial-mass star. We derive the best-fit distance of 0.46 kpc by fitting the stellar luminosity (as a function of the distance and effective temperature of the central star) with the adopted post-AGB evolutionary tracks. Our excitation energy diagram analysis indicate high excitation temperatures in the photodissociation region (PDR) beyond the ionized part of the nebula, suggesting extra heating by shock interactions between the slow AGB wind and the fast PN wind. Through iterative fitting using the Cloudy code with empirically-derived constraints, we find the best-fit dusty photoionization model of the object that would inclusively reproduce all of the adopted panchromatic observational data. The estimated total gas mass (0.41 Msun) corresponds to the mass ejected during the last AGB thermal pulse event predicted for a 2.5 Msun initial-mass star. A significant fraction of the total mass (about 70 percent) is found to exist in the PDR, demonstrating the critical importance of the PDR in PNe that are generally recognized as the hallmark of ionized/H+ regions.Comment: 34 pages, 13 Figures and 16 Tables, accepted for publication in the Astrophysical Journal Supplement Serie

    MicroRNAs and Drinking: Association between the Pre-miR-27a rs895819 Polymorphism and Alcohol Consumption in a Mediterranean Population

    Recently, microRNAs (miRNA) have been proposed as regulators in the different processes involved in alcohol intake, and differences have been found in the miRNA expression profile in alcoholics. However, no study has focused on analyzing polymorphisms in genes encoding miRNAs and daily alcohol consumption at the population level. Our aim was to investigate the association between a functional polymorphism in the pre-miR-27a (rs895819 A>G) gene and alcohol consumption in an elderly population. We undertook a cross-sectional study of PREvencion con DIeta MEDiterranea (PREDIMED)-Valencia participants (n = 1007, including men and women aged 67 +/- 7 years) and measured their alcohol consumption (total and alcoholic beverages) through a validated questionnaire. We found a strong association between the pre-miR-27a polymorphism and total alcohol intake, this being higher in GG subjects (5.2 +/- 0.4 in AA, 5.9 +/- 0.5 in AG and 9.1 +/- 1.8 g/day in GG; p(adjusted) = 0.019). We also found a statistically-significant association of the pre-miR-27a polymorphism with the risk of having a high alcohol intake (> 2 drinks/day in men and > 1 in women): 5.9\% in AA versus 17.5\% in GG; p(adjusted) < 0.001. In the sensitivity analysis, this association was homogeneous for sex, obesity and Mediterranean diet adherence.     Oral lichen planus and its relationship with systemic diseases. A review of evidence

    Oral lichen planus (OLP) is one of the most common dermatological diseases which are present in the oral cavity. It is a chronic autoimmune, mucocutaneous disease that affects the oral mucosa as well as the skin, genital mucosa and other sites. Review the relevant information to OLP and its relationship with systemic diseases. Searches were carried out in the Medline/PubMed, Lilacs, Bireme, BVS, and SciELO databases by using key-words. After an initial search that provided us with 243 papers, this number was reduced to 78 from the last seven years. One of the first criteria adopted was a selective reading of the abstracts of articles for the elimination of publications that presented less information regarding the subject proposed for this work. All the selected articles were read in their entirety by all of the authors, who came to a consensus about their level of evidence. The Scottish Intercollegiate Guidelines Network (SIGN) criteria were used as the criteria of methodological validation. Only 9 articles showed an evidence level of 1+, 2+, 3 or 4, as well as a recommendation level of A, B, C or D. Three of them were non-systematic reviews, one was a cohort study and only one was a controlled clinical trial. Three of the studies were case series, with respective sample sizes of 45, 171 and 633 patients. Several factors have been associated with OLP. Patients with OLP are carriers of a disease with systemic implications and may need the care of a multidisciplinary team. The correct diagnosis of any pathology is critical to making effective treatment and minimizes iatrogenic harm. For OLP is no different, taking into account its association with numerous systemic diseases that require special attention from health professionals. Periodic follow-up of all patients with OLP is recommended

    Understanding Differences in the Crystallization Kinetics between One-Step Slot-Die Coating and Spin Coating of MAPbI₃ Using Multimodal In Situ Optical Spectroscopy

    To develop a detailed understanding about halide perovskite processing from solution, the crystallization processes are investigated during spin coating and slot-die coating of MAPbI3 at different evaporation rates by simultaneous in situ photoluminescence, light scattering, and absorption measurements. Based on the time evolution of the optical parameters it is found that for both processing methods initially solvent-complex-structures form, followed by perovskite crystallization. The latter proceeds in two stages for spin coating, while for slot-die coating only one perovskite crystallization phase occurs. For both processing methods, it is found that with increasing evaporation rates, the crystallization kinetics of the solvent-complex structure and the perovskite crystallization remain constant on a relative time scale, whereas the duration of the second perovskite crystallization in spin coating increases. This second perovskite crystallization appears restricted due to differences in solvent-complex phase morphologies from which the perovskite forms. The work emphasizes the importance of the exact precursor state properties on the perovskite formation. It further demonstrates that detailed analyses of multimodal optical in situ spectroscopy allows gaining a fundamental understanding of the crystallization processes that take place during solution processing of halide perovskites, independent from the specific processing method
